چکیده

The presently used heterotrophic plate count (HPC) for the evaluation of the total number of bacteria in a sample of drinking water takes 7 days of incubation. When the results are known, the water has been consumed and is ineffective for the protection of the health of the consumers. Operators of water treatment systems need to know the bacterial water quality in near real time. Contamination of the system, whether it is intentional, accidental, or due to an inadequate disinfectant residual needs to be discovered much sooner because intervention can then take place in the form of flushing low quality water and/or raising the disinfectant residual. The purpose of this study was therefore to determine if a rapid ATP assay can estimate the HPC in minutes. Two additional methods were used for some samples. The first method was the acridine orange direct count (AODC) that enumerates both viable and nonviable bacteria. The second method was the direct viable count (DVC) that enumerates only viable bacteria. Water samples were obtained from local, national, and international locations. The sample selection criteria were based on proximity to the laboratory, cooperating water utilities, and the travel of the authors. The results of the study show that the rapid ATP assay is highly correlated with the conventional plate count method and the DVC method. The significance of the ATP assay is that it can determine the bacterial quality of the drinking water in less than 5 min. C © 2001 John Wiley & Sons, Inc. Field Analyt Chem Technol 5: 185–189, 2001

A Rapid Method for Detecting Bacteria in Drinking Water

A rapid determination of the total bacterial count in drinking water is important to the operators of treatment plants and distribution systems. It will allow corrective measures in real-time, such as increasing the disinfectant dose or removing water that has high bacterial numbers. The present heterotrophic plate count (HPC) analysis takes seven days and is not useful for operational interven...

Cultivation - independent assessment of viability with flow cytometry

Less than 1 % of bacteria in drinking water are typically culturable on conventional growth media with the heterotrophic plate count (HPC) method. In addition, it is believed that some culturable bacteria can enter a state of unculturability when exposed to nutrient-poor environments. Therefore, a need exists for rapid and cultivation-free methods with which the viability/activity of drinking w...

Detection and Persistence of Clinical Escherichia coli in Drinking Water Evaluated by a Rapid Enzyme Assay and qPCR

The aims of this study were to evaluate two methods, qPCR and a chemiluminescent assay (ColiLight II), for rapid detection of E. coli in water, and to examine the survival and persistence of clinical E. coli in drinking water and biofilm using qPCR and ColiLight II. qPCR and ColiLight II were compared with a cultivation-based method (MPN), and survival and persistence of four clinical E. coli s...

Expression and Purification of the luciferase enzyme and in Vivo ATP Assay

Introduction: Gene expression and purification of luciferases from the firefly, Lampyris turkestanicus, and optimization of cellular ATP measurements were performed. Methods: cDNA encoding luciferases from Lampyris turkestanicus was transferred from pQE30 vector into pET28a expression vector and pLtu28 was built. Newly constructed vector was expressed in E. coli XL1 Blue and the recombinant l...

Rapid Monitoring of Indicator Coliforms in Drinking Water by an Enzymatic Assay

Coliform group has been extensively used as an indicator of drinking water quality and historically led to the public health protection concept. Multiple tube fermentation technique has been currently used for assessment of the microbial quality of drinking water. This method, however, has limitations. Enzymatic assay constitute an alternative approach for detecting indicator bacteria, namely t...
